Real time measurement of myocardial substrate selection in vivo using hyperpolarized 13C magnetic resonance
نویسندگان
چکیده
Background Cardiac dysfunction is often associated with a shift in substrate preference and metabolism, but current in vivo techniques only provide information on substrate uptake. Hyperpolarized (HP) MR has the unique ability to detect metabolism in vivo and is highly specific. To evaluate the prospects for measuring myocardial substrate selection in vivo, HP [1-C]pyruvate and [1-C] butyrate were co-infused into rats.

متن کاملClinical Implications of Cardiac Hyperpolarized Magnetic Resonance Imaging
Alterations in cardiac metabolism are now considered a cause, rather than a result, of cardiac disease. Although magnetic resonance spectroscopy has allowed investigation of myocardial energetics, the inherently low sensitivity of the technique has limited its clinical application in the study of cardiac metabolism. The development of a novel hyperpolarization technique, based on the process of...
متن کاملHyperpolarized 13C and 31P magnetic resonance spectroscopy identify pyruvate dehydrogenase as a therapeutic target in obesity cardiomyopathy
Background Although the link between altered myocardial substrate selection and impaired function in obesity cardiomyopathy is unclear, it is likely to involve altered activity of pyruvate dehydrogenase (PDH), a key regulator of carbohydrate metabolism. We hypothesised that; 1) obesity would impair myocardial carbohydrate oxidation, in turn reducing energetic reserve and predisposing to functio...
متن کاملIncreasing Pyruvate Dehydrogenase Flux as a Treatment for Diabetic Cardiomyopathy: A Combined 13C Hyperpolarized Magnetic Resonance and Echocardiography Study.
Although diabetic cardiomyopathy is widely recognized, there are no specific treatments available. Altered myocardial substrate selection has emerged as a candidate mechanism behind the development of cardiac dysfunction in diabetes. As pyruvate dehydrogenase (PDH) activity appears central to the balance of substrate use, we aimed to investigate the relationship between PDH flux and myocardial ...
متن کاملHyperpolarized 13C urea myocardial first-pass perfusion imaging using velocity-selective excitation
BACKGROUND A velocity-selective binomial excitation scheme for myocardial first-pass perfusion measurements with hyperpolarized 13C substrates, which preserves bolus magnetization inside the blood pool, is presented. The proposed method is evaluated against gadolinium-enhanced 1H measurements in-vivo. METHODS The proposed excitation with an echo-planar imaging readout was implemented on a cli...
